TitleInflammatory molecules in aqueous humor of children with endogenous uveitis

PurposeTo investigate immune mediator profile in the aqueous humor (AqH) of children with endogenous uveitis (EU) and its relationships with disease location, activity and presence of secondary glaucoma.
MethodsConcentrations of 45 inflammatory molecules were measured using multiplex bead immunoassay (xMAP, Luminex, USA; cytokine/chemokine/growth factor 45-plex human panel, ProcartaPlex, eBioscience, Austria) and TGF-β1, TGF-β2 using enzyme-linked immunosorbent assay (ELISA) (Bender Medsystems, Austria) in 43 AqH samples obtained during cataract surgery from 40 children aged 5 – 15 years with anterior (32 eyes), intermediate (8), posterior (1) or panuveitis (2). Remission of uveitis was in 27 eyes, mild activity in 16.
ResultsFrequency of detection of molecules varied and was highest (70 – 100%) for MCP-1, MIP-1α, MIP-1β, Eotaxin, IP-10, VEGF-A, TGF-β1, TGF-β2, SDF-1α, HGF, SCF, IL-7, IL-6 and LIF. Levels of IL-2 (p=0.038), Eotaxin (p=0.02), TGF-beta2 (p=0.008) were higher in anterior than in intermediate uveitis. Levels of IL-10 (p=0.03) were higher, IL-2 (p=0.017) and EGF (p=0,003) lower in remission than in active uveitis. Higher levels of IL-7 (p=0.017) and tendency to higher levels MCP-1 (p=0.064) and lower levels Eotaxin (p= 0,059) and IL-15 (p=0,075) were found in patients with than in patients without uveitic glaucoma.
ConclusionMultiple inflammatory molecules, detected in AqH of children with EU, may play pathogenic role in the disease and serve as targets for development of treatment.
