TitleKiller cell Immunoglobulin-like Receptors and ligands in patients with CMV-Anterior Uveitis

PurposeCytomegalovirus Anterior Uveitis(CMV-AU)manifests in immunocompetent individuals of which the mechanisms are unknown. In this study, we have investigated the role of NK cells in CMV-AU. Important determinants that differentiate human NK responses are the Killer Cell Immunoglobulin-like Receptors (KIR) and their ligands, HLA class I subgroups. Combinations of polymorphisms of KIR and HLA are known to be associated with resistance against virus infection and progression.
MethodsKIR ligands in 118 Chinese Singaporean CMV-AU cases were determined using Next Generation Sequencing (NGS). KIR genotypes were determined using a PCR-SSP method. The data was compared against the profiles of the healthy Chinese Singaporean population.
ResultsAmong HLA-B Bw4 allele groups (80Ile, 80Thr), the frequency of the 80Ile allele group was significantly increased and the frequency of 80Thr allele group was significantly lower in CMV-AU patients compared to those in healthy controls. No difference was observed in the frequencies of ligands for KIR2DL1/2DL3 and the group A haplotype.
ConclusionGenetic profiles of KIRs and their ligands in CMV-AU contrast to the KIR/HLA associations reported previously for symptomatic systemic primary CMV infection, implying that the immunological mechanisms against CMV are likely distinct.
