TitleCirculating microRNA alterations in serum during the development of experimental autoimmune uveoretinitis
PurposeTo investigated the associations between the development of experimental autoimmune uveoretinitis (EAU) in rat and the alterations in the circulating microRNA (miRNA) in serum.
MethodsFor induction of EAU, Lewis rats were immunized with interphotoreceptor retinoid binding protein (IRBP) peptide (R14) and clinical score of EAU was assessed on days 0 (baseline), 7, 14, and 21. Total RNA was isolated from serum on days 0, 7, 14, and 21 and used for microarray analysis. The levels of miRNA in serum was compared between baseline (day 0) and day 7, 14, 21.
ResultsThe mean clinical score of EAU was 0 (day 7), 4.3 (day 14), and 0.7 (day 21). A miRNA-microarray analysis showed significantly increased expression of 6 (day 7), 12 (day 14), and 10 (day 21) miRNAs, whereas 22 (day 7), 22 (day 14), and 21 (day 21) miRNAs were significantly reduced following EAU development. In particular, miRNA-146a-5p and miRNA-150-5p were significantly elevated in serum on peak stage (day 14) and resolution stage (day 21) of EAU. Bioinformatics analysis revealed that TGF-beta signaling pathway and mucin type O-glycan biosynthesis were the topmost commonly significant targeted pathways during EAU development.
ConclusionThe miRNA signatures in serum may provide the rationale for the development of new treatment strategy or could be served as biomarker for assessment of the activity of non-infectious uveitis.
