Development of transparent triamcinolone acetonide solution for intravitreal administration for uveitis treatments
Purpose
Intravitreal corticosteroid therapy can be useful for certain types of noninfectious uveitis that involve the posterior segment of the eye. However, triamcinolone acetonide (TA) is a low water soluble suspension. It remains as white crystalline deposits in the vitreous and can cause visual disturbances. The aim of our research was to evaluate the efficacy of intravitreal triamcinolone acetonide in conjunction with hyaluronic acid (HA) for the treatment of uveitis which involved the posterior segment.
Methods
TA is modified by amino acid using esterification, and the HA-based conjugates was synthesized from precursor using the carbodiimide coupling agent. The structure was identified by 1H-NMR spectroscopy. After the purification and characterization, the anti-inflammatory effect was evaluated in the endotoxin-induced uveitis (EIU) animal model. EIU was induced by a single intravitreal injection of LPS using a Hamilton microinjector under a dissecting microscope.
Results
The designed HA-TA coupling reaction reveals >98 % conjugation efficiency in the dimethyl sulfoxide/water system. The HA-TA conjugates is water soluble and transparent which is different from the commercial TA suspension. The cell number and protein in aqueous humor was significant decreased in EIU model.
Conclusion
HA-TA was successfully synthesized and revealed transparent character. Potential treatment effects of HA-TA conjugates were observed in EIU animal model.
