TitleEffectiveness of adalimumab biosimilars in patients with non-infectious uveitis: a monocentric observational study of non-medical switch from adalimumab originator to ABP501 biosimilar and from ABP501 biosimilar to SB5 biosimilar.

PurposeTo evaluate the clinical efficacy of non-medical biosimilar(BS)switch of adalimumab in preventing relapses of non-infectious uveitis(NIU)and any differences respect to the originator.To date,this is the first study to include NIUpatients who performed a double switch of adalimumab BSs
MethodsThis retrospective study included19 NIU patients initially treated with the adalimumab originator for one year(T0), followed by ABP501 BS for the second year (T1) and SB5 BS for the third year (T2). Best-corrected visual acuity (VA), relapse rates, total steroid and other immunosuppressants dosage,number of intravitreal steroid implants,and complications were evaluated
Results19patients were included,9 female (47.4%). A significant difference in corticosteroid usage across the3 years was observed (p=0.027).Specifically, corticosteroid dosage was higher during T0 andT2 than T1(p=0.023).The dosage of other immunosuppressants was lower duringT2, though this was not statistically significant.No significant differences were observed in ocular or systemic recurrence rates (p=0.7768 for T1 vs T0, p=0.0719 for T2 vs T1) or the number of dexamethasone implants. A significant correlation existed between steroid dosage atT0 and relapses at T0(rho=0.5297,p=0.0197).No significant correlations were found between VA,steroid dosage,or disease duration
ConclusionSwitching from adalimumab originator to ABP501 and SB5 BSs did not significantly affect recurrence rates,complications,or VA in NIUpatients. No significant differences in efficacy or safety profiles were observed between the three therapies.
